Tissue Culture of Cyclamen persicum Mill.

With leaf，petiole and stem segments of cyclamen as explants，using different types of disinfectants and disinfection time，disinfection of cyclamen explant was studied.Induction effect of cyclamen callus was studied respectively from explant type，basic culture medium type，and hormone allocated proportion between 6-BA and NAA.Best conditions of cyclamen cluster bud proliferation were chose by multi factor orthogonal test.Role of carbon on cyclamen rooting was researched.Results showed that disinfection effect of sodium hypochlorite was better than mercuric chloride.The best disinfection time was 10 min for explant soaked with 2% sodium hypochlorite.Leaf as explant was easier to induce callus.Basic culture medium for callus induction was MS.Medium for callus induction and bud differentiation was MS+6-BA1.0 mgL+NAA0.2 mgL.Medium for shoot proliferation was 12 MS+6-BA 1.0 mgL+NAA 0.2 mgL.Medium for rooting was 12 MS+NAA0.2 mgL+3% sucrose+AGAR 7 gL+activated carbon 0.5 gL.
