Micropropagation Technique of Ethnic Medicinal Plant Ardisia gigantifolia Stapf

  • Effects of different plant growth regulators,illumination intensity,pH and additional nutrient on induce,proliferation and rooting of adventitious buds were studied by taking stem segment of Ardisia gigantifolia Stapf as explant.Results showed that the optimal proliferation medium for Ardisia gigantifolia Stapf was WPM with 2.0 mg/L BAP、0.2 mg/L NAA and 0.2 mg/L 2,4-D.The optimal pH was 6.0,plantlets grew well when illumination intensity was 100 μmol/(m·s).1 g/L peptone,50 g/L coconut juice and 50 g/L banana were good for cultivating strong seedling,and 50 g/L banana was the best.The best rooting medium was MS medium supplemented with 2.0 mg/L IBA and 0.5 mg/L NAA,rooting rate reached 93.3%,Ardisia gigantifolia Stapf grew well with a survival rate of 95% when transplanted into mixed matrix of coconut tree branny:river sand=2∶1.
