冷冻丙酮法分离杜仲子油中α-亚麻酸工艺
Separation of α-Linolenic Acid from Eucommia Seed Oil by Freezing Acetone Method

摘要: 采用响应面法优化冷冻丙酮法分离杜仲子油中α-亚麻酸的工艺条件。在单因素试验基础上,选取冷冻温度、V(丙酮)/m(脂肪酸)、冷冻时间为自变量,以α-亚麻酸纯度为响应值,对工艺条件进行优化,并通过气相色谱-质谱联用技术分析杜仲子油中α-亚麻酸的含量。结果表明:在冷冻温度-44 ℃,冷冻时间3.3 h,V(丙酮)/m(脂肪酸)=4.2条件下,杜仲子油中α-亚麻酸纯度为28.05 mg/mL,其含量达到80.11%,杜仲子油中α-亚麻酸的含量由60.14%提高到80.11%。Abstract: Response surface analysis method was applied to optimize the enrichment of α-Linolenic acid by freezing acetone method from Eucommia seed oil.According to the single factor experiments,optimizeing freezing temperature,V(acetone)/m(fattyacid)and freezing time by using Box-Behnken experiment design to optimize.And the content of α-Linolenic acid could be analyzed by GC-MS.The optimal conditions were determined as follows:freezing temperature -44 ℃,V(acetone)/m(fattyacid)4.2,freezing time 3.3 h.The actual α-Linolenic acid purity was 28.05 mg/mL and its content was 80.11%,compared to the theoretical value,the relative error of 0.43%.The content of α-Linolenic acid was improved from 60.14% to 80.11%.
