响应面法优化微波提取冬枣叶总黄酮及其抗氧化活性

Optimization of Microwave Extraction of Total Tlavones from‘Dongzao’Jujube Leaves by Response Surface Methodology and Flavones Antioxidant Activity

  • 摘要: 采用微波法提取冬枣叶中总黄酮,研究了微波温度、微波时间、微波功率、液料比和乙醇体积分数对总黄酮提取率的影响。在单因素试验的基础上,采用Box-Behnken试验设计及响应面分析法对冬枣叶总黄酮的微波提取工艺进行优化,建立回归方程。得最佳提取工艺条件:微波温度70 ℃、微波时间6 min、微波功率500 W、液料比60∶1、乙醇体积分数50%,在此条件下,总黄酮提取率为3.760%,与响应面拟合所得方程的预测值3.757%符合良好。对提取的总黄酮抗氧化性研究表明,冬枣叶总黄酮对于羟基自由基清除能力较强,有一定的抗氧化活性。

     

    Abstract: Microwave was used to extract total flavones from‘Dongzao’jujube leaves.Effects of the extraction parameters on the yield of total flavones from‘Dongzao’jujube leaves were studied,such as microwave temperature,microwave time, microwave power,liquid-solid ratio and ethanol concentration.Box-Behnken design and response surface analysis were adopted to establish regression equation of extraction of total flavones from‘Dongzao’jujube leaves on single factor tests.Optimal extraction conditions were microwave temperature 70 ℃,microwave time 6min,microwave power 500 W,liquid-solid ratio 60∶1 and ethanol concentration 50%.Under the conditions,the yield of total flavones was 3.760%,which was consistent with the predicted yield of total flavones of 3.757%.Results showed that the total flavones from leaves of‘Dongzao’jujube were studied.Results also showed that the total flavones had strong activity for hydroxyl radical scavenging,and had a good antioxidant activity.
