柑橘脱毒快繁及微芽嫁接技术试验研究

Test and Research on Virus-free Rapid Propagation Technology and Micro Bud Grafting Technology of Citrus

  • 摘要: 以红江橙、沙糖桔的顶芽、侧芽和茎段为外植体,通过诱导培养形成脱毒不定芽接穗;通过微芽嫁接于砧木实生苗,培养成完整的无毒植株。试验结果表明:外植体灭菌处理成功率达86%,诱导不定芽的最适培养基为MT+6-BA 3.0 mg/L+IBA 0.1 mg/L+KT 0.1 mg/L;不定芽继代增殖的最适培养基为MT+6-BA 2.0 mg/L+IBA 0.1 mg/L,增殖系数为2.3;砧木种子播种成苗率达90%;微芽嫁接愈合成活率平均达87%。该研究为柑橘脱毒种苗提供了一种新型快速的繁殖方式。

     

    Abstract: Using Citrus sinensis Osbeck cv.Hongjiang and Citrus Shatangju,virus-free adventitious buds were formed by inducing culture.Complete non-toxic plants were cultured by grafting seedling to rootstock seedling.Test results showed that success rate of explant sterilization was 86%,optimum medium of induction of adventitious buds was MT+6-BA 3.0 mg/L+ IBA 0.1 mg/L+KT 0.1 mg/L.Optimum medium of adventitious bud subculture was MT+6-BA 2.0 mg/L+IBA 0.1 mg/L and proliferation coefficient was 2.3.Seedling rate of stock seed reached 90%.Average survival rate of micrograft reached 87%.This study provided a new way of rapid propagation for Citrus virus-free seedling.
